Coumarin indole hybrid pharmacophores in medicinal chemistry design synthesis biological activities and structure activity relationships

Abstract

Coumarin-indole hybrid pharmacophores have emerged as promising multifunctional scaffolds in medicinal chemistry by integrating two privileged heterocyclic frameworks with complementary pharmacological properties. This review systematically summarizes recent advances in the design strategies, synthetic approaches, biological activities, and structure-activity relationships (SAR) of coumarin-indole hybrids. A structured literature search was conducted using PubMed, Scopus, Web of Science, and Google Scholar to identify relevant studies published up to March 2026, with emphasis on original research articles and key review papers describing the synthesis, biological evaluation, and pharmacological applications of these hybrid molecules. The available evidence demonstrates that coumarin-indole hybrids exhibit remarkable anticancer, antimicrobial, antiviral, anti-inflammatory, neuroprotective, enzyme inhibitory, antidiabetic, antileishmanial, and antihyperlipidemic activities through multiple mechanisms, including tubulin inhibition, apoptosis induction, enzyme modulation, and multi-target pathway regulation. Comparative analysis of representative hybrids reveals that substitution patterns, linker architecture, and electronic effects play critical roles in determining biological activity, providing valuable SAR insights for the rational optimization of these compounds. This review further discusses pharmacokinetic considerations, ADMET (absorption, distribution, metabolism, excretion, and toxicity) challenges, opportunities for clinical translation, and emerging trends such as artificial intelligence-assisted drug design and multi-target-directed ligand development. By integrating current knowledge on design strategies, representative biological studies, SAR trends, pharmacokinetic considerations, and future perspectives, this review provides a comprehensive and up-to-date resource that supports the rational development of next-generation coumarin-indole hybrid therapeutics.
